Abstract
Infra-Red (IR) analysis has been long accepted as a powerful tool in the understanding and characterization of proteins. IR provides a data rich insight into the folding of proteins, and in particular the Amide I (~1600 – 1700 cm-1) region gives detailed secondary structural composition information. Typically, traditional Fourier Transform IR (FTIR) is used, but high concentration requirements, low repeatability, water subtraction issues, drift and limited automation have prevented this technology from reaching its full potential. Likewise, Circular Dichroism also provides a very complementary tool to FTIR, but data interpretation is subjective. With concentration limits required to be below <0.5 mg/ml, and incompatibility with complex buffers, the presence of surfactants, excipients and reducing agents. Often prevents high quality data from being obtained, or the data generated being subjective. Microfluidic Modulation Spectroscopy (MMS) is a new key technology that focuses on the IR Amide I region to produce exceptionally high data quality and with extraordinarily high data reproducibility, and it is compatible with a very broad concentration range( >0.1 - >200 mg/ml) and with complex buffer systems and excipients. This poster presents the technical benefits of MMS and its application in the protein structural workflow.
