IR Spectral Signatures of GC, AU, and GU Base Pairing in H2O-based Buffer by MMS

Introduction

RNA structure emerges from the cumulative effects of Watson-Crick and non-Watson-Crick base pairing, π-stacking interactions within and between RNA strands, and solvent interactions, resulting in diverse functional conformations. While traditional biophysical techniques such as ultraviolet (UV) melting curves and circular dichroism (CD) spectroscopy provide valuable structural and thermodynamic insights, they generally do not resolve specific base pair types or populations. Nuclear magnetic resonance (NMR) spectroscopy can reveal detailed structural insights, but requires high sample concentrations, imposes intrinsic size limitations, and has significant constraints on instrument throughput and accessibility.
