Poster: Probing RNA-based Pairing and Ligand Interactions with Infrared-based Microphiletic Modulation Spectroscopy

Abstract

RNA structure and dynamics are essential for understanding its biological functions, stability, and potential as both a therapeutic agent and target. This study explores Microfluidic Modulation Spectroscopy (MMS)—a cutting-edge infrared spectroscopy technique capable of detecting subtle changes in RNA conformation and hydrogen bonding patterns. Using the AuroraTX system from RedShiftBio, we demonstrate MMS’s ability to analyze RNA structural elements, including base pairing, secondary structures like i-motifs and G-quadruplexes, and thermal melting dynamics. Additionally, MMS proves effective in detecting ligand-induced RNA conformational changes, showcasing its potential as a powerful tool for RNA-targeted drug discovery. These findings establish MMS as a valuable technique for advancing RNA structural research and therapeutic development.
